OECD 456: H295R Steroidogenesis assay
This assay provides an in vitro screen for identifying substances that interfere with steroidogenesis, specifically, production of 17ß-estradiol and testosterone and is suitable for Level 2 endocrine disruption testing.
The human H295R adreno-carcinoma cell line used for the assay expresses genes that encode for all the key enzymes for steroidogenesis. Following exposure to a range of concentrations of the test substrate, concentrations of hormones in the cell medium are measured and compared with fixed concentrations of a known inducer and inhibitor of hormone production.
